Abstract
To develop an efficient tissue culture protocol for callus induction and
regeneration in two popular indica cultivars i.e. IR 64 and Swarna was
undertaken. The explants used for initiation of culture were mature seeds. For
callus induction in Swarna and IR 64, MS (Murashige and Skoog, 1962) media
was used with different concentration of 2, 4-D, Kinetin, L-proline and Casine
hydrolysate in 10 combinations. Best callus induction in terms of friability and
growth in Swarna were obtained in the medium having combinations of MS
media supplemented with 2, 4-D (2 mg/ l) + Kinetin (0.4 mg/ l) + L-proline (500
mg/ l) + Casein hydrolysate (300 mg/ l) + maltose (30 g/ l) + agar-agar (0.8 %
w/v) with 87 % callus induction percentage, whereas in IR-64, it was found best
with MS media supplemented with 2, 4-D (2 mg/ l) + L-proline (500 mg/ l) +
Casein hydrolysate (300 mg/l) + maltose (30 g/ l) + agar-agar (0.8 % w/v) with 78
% callus induction percentage.
