Abstract
The present investigation entitled “Optimization of Micropropagation Techniques in Spine Gourd by using Response Surface Methodology” was conducted with the objective to optimize the various levels and combinations of media supplements on culture media for amount of callus growth on different explants viz., leaf, petiole, internode and to standardize the protocol for in-vitro regeneration of female Momodica dioica genotype RMDSG-2 using nodal and shoot tip explants. D-optimal response surface methodology was used for experimental designing and to analyze the generated data for the effect of various levels of media supplements on the node and shoot tip explant at initiation, multiplication and at rooting stages. MS media was used as a basal media and was supplemented with various levels of media supplements as suggested by RSM. The best optimized combinations observed from initiation stage for each experiment were again compared in CRD and statistically observed best treatment was used for multiplication of shoots from nodal as well as shoot tip explants. The best regenerated plants were then transplanted for rooting study. Among the leaf, petiole and internode explants, the leaf explants showed earliest callus initiation with high amount of callus on a MS media supplemented with 1.0 and 2.0 mgl-1 NAA. The white fragile callus was observed in the all treatments except 2.0 mgl-1 TDZ + 1.5 mgl-1 BAP (brown fragile callus) and 1.5 mgl-1 BAP + 1.0 mgl-1 Kinetin (pink fragile callus).
