Screening of Brinjal Genotypes of North East India Against Bacterial Wilt Using SSR Markers

Abstract
Genetic variability and correlation were studied and the characters like fruit weight (g), number of fruits per plant had high PCV and GCV whereas, heritability was observed high for all the characters. Genetic advance was highest for fruit weight (229.06%). Correlation study indicated that fruit yield per plant was positively correlated with fruit weight. Maximum inter cluster distance was observed between cluster III and V indicated the genotypes within these were highly divergent. For screening of bacterial wilt severity, days to first incidence of bacterial wilt symptoms, plant disease incidence at 15 DAI, 25 DAI, 30 DAI, 50 DAI, 60 DAI, and disease severity index were recorded. Disease severity was found lowest in CHFB- 33, followed by CHFB-29 and CHFB-22. On the basis of screening in field condition, the genotypes CHFB- 22 , CHFB-29, CHFB-33 were found resistant, CHFB- 21 , CHFB-23 , CHFB-24, CHFB- 26, CHFB-28, CHFB- 30, CHFB-31, CHFB- 34, CHFB-35 were found moderately resistant, CHFB-20, CHFB-25, CHFB-18, CHFB-19 moderately susceptible, CHFB- 37, CHFB- 27, CHFB-32 susceptible and, CHFB-36 highly susceptible against bacterial wilt of brinjal. DNA extraction was done by CTAB method and PCR using 20 SSR markers. Primer emb01J19 showed highest polymorphic information content, diversity index and matrix index. In the study of molecular diversity, genotypes were grouped broadly into two major clusters I and II, where cluster I is further divided into sub clusters IA and IB using SSR markers. The sub cluster IA again divided into two sub clusters, IA1 and IA2 on molecular basis. All the genotypes grouped under above clusters considered in two main clusters on the basis of its diversity, resistant and susceptibility. The genotypes Arka Nidhi and Arka Keshav were recorded to be highly resistant which falls under sub cluster IA. The genotypes CHFB-22, CHFB-26, CHFB-29, CHFB-21, CHFB-24, CHFB- 23, CHFB-31, CHFB-26, CHFB-28, CHFB-30, CHFB-33, CHFB-34, CHFB-35, CHFB-19, CHFB-18, CHFB-37, CHFB-20, CHFB-25, CHFB-36 observed resistant to moderately susceptible under major cluster I while, CHFB-27 and CHFB-32 grouped under major cluster II showed susceptibility through SSR markers. Athough genotype CHFB-36 observed most susceptible in field condition, it was grouped under major cluster I in molecular analysis due to the similar genetic makeup of the genotype with other genotypes of major cluster I. It is concluded that genotypes CHFB-22 and CHFB-29 found nearer to resistant check variety Arka Nidhi and Arka Keshav under major cluster I and also CHFB-33 appeared separately in major cluster I sub cluster IA- IA1 grouped under resistant and these three lines can be utilized for improvement through breeding.

An experiment entitled “Screening of Brinjal Genotypes of North East India against Bacterial Wilt Using SSR Markers” was conducted at polyhouse of Vegetable Research Farm, Deptt. of Vegetable Science, College of Horticulture and Forestry, CAU, Pasighat, Arunachal Pradesh during the year 2019-20 with 22 brinjal genotypes in Randomised Block Design with three replications.
