Abstract
The present investigati on was undertaken for study of introgress ion of gene from wild specie to culti vated species of pigeonpea (cajanus cajan) and to study simi larity of hybrid with parents.CT AB method of DNA isolation procedure was standardized to yield good quality of DNA from pigeonpea. PCR optimi za ti on with appropriate concentration as well as temperature and time adjustment was done. Decamer primer used to analyze hybridization study with DNA. The products were run on agarose gel and photographed.ln present investigati on random amplifi ed polymorphic DNA (RAPD) marker was used to study the hybridity of eight crosses of pigeonpea (cajanus cajan). A total 12 primers were screened out of which 6 gave good results. Amplification of genomic DNA of 8 crosses , using RAPD primers yielded 306 fragments that could be scored, of which 144 were polymorphic in all crosses. Percent of polymorphism ranged from I2.5 to I 00 percent. There were seven types on the basis of banding pattern shows in I 51 type both the parents and offspring's contained bands. In remaining 6 groups either female or male or F 1 or both parents or one parent and F 1 contain bands. In the SSR results, out of 20 primers 6 gave good results. But all primers have given monomorphic bands, so we cant distingui sh relationship between parents and hybrid . Similarity matrix obtained through NT-SYS PC software revealed that male is more simil ar to Fl as compared to fema le. Present investigation indicate that for hybrid conformation RAPD technique is useful.
