Abstract
RT-PCR technique had been chosen to study the caprine antigen presenting cells induced Th cells cytokine secretion profile. Macrophages induced more IFN- γ γ γ γ γ than IL-4 cytokine mRNA and in contrast, B cells induced higher IL-4 production than IFN-γ γ γ γ γ. In general, IL-4 mRNAs were more in two antigen pulsed cultures as assessed by integrated densitometry analysis. M. paratuberculosis induced higher IFN-γ γ γ γ γ mRNAs than S. aureus. Dexamethasone (50μg/ml) increased the IL-4 production than indomethacin. Thus, RT-PCR based assay system for the detection of cytokine is a fast, economical method to assess simultaneously several cytokines during an immune response. But further studies are needed to develop a competitive RT-PCR assay system to quantify the cytokines secreted by caprine effector cells at various stages of infection.
