RAPD-PCR CHARACTERIZATION OF CAMPYLOBACT ER JEJUNI ISOLATES OBTAINED FROM RAW MILK SAMPLES IN CHENNAI

Abstract
A study was conducted to investigate strain variations among 42 C. jejuni iso—lates obtained from raw milk by Randomly amplified polymorphic DNA (RAPD) method two randomly designed primer of 10-mers .RAPD profiles were generated with one primer(7718) and the other one(129O) was unable to do so.The resultant finger prints comprised between 7 and 16 bands for Cjejuni isolates with sizes between 70 bp and 2400 bp able to amplify C. jejuni DNA .The 13 distinct banding profiles generated by RAPD were analyzed using computer software and the prepared dendrogram has grouped the isolates in to four major clusters. The RAPD assay has been proved to have excellent discrimination ability due to the fact that the entire genome is the target in genotyping . It can be considered the most advantageous assay in terms of discrimination ability, cost and ease.
