Abstract
The present investigation on “REGENERATION STUDY IN
BRINJAL (Solanum melongena L.)” was undertaken to identify suitable
explants and media for micropropagation. The seeds of Solanum melongena
were collected from Vegetable project, Department of Horticulture, MPKV,
Rahuri, Dist. Ahmednagar. Various experiments were conducted to study
different aspects of micropropagation. Sterilization with 0.1 % HgCl2 for 5-6
min. duration proved to be the best and gave 80 percent aseptic culture
establishment. Cotyledonary leaf, shoot tip and nodal segment showed
positive response for establishment and multiplication.
Different media concentrations were tried for establishment of
culture, the media MS+BAP 2.0 mg/l+ Kinetin 1.0 mg/l gave maximum
percent establishment i.e.100 % for cotyledonary leaf, 96.66% for shoot tip
and 93.33 % for nodal segment. MS + BAP 2.0 mg/l+ Kinetin 1.0 mg/l proved superior for
establishment and required least days for establishment i.e. 9.33 days for
cotyledonary leaf, 9.33 days for shoot tip and 9.66 days for nodal segment.
The media MS + BAP 2.0 mg/l +Kinetin 1.0 gave the maximum
number of multiple shoots per culture i.e. 6.66, 5.33 and 4.33 to
cotyledonary leaf, shoot tip and nodal segment respectively after
establishment of explants.
The optimums rooting to the shoot were recorded on MS
supplemented with 0.5 mg/l IBA. All rooted shoots were established after
hardening.
The regeneration protocol for brinjal (Manjari gota) has been
standardized from establishment of explants to rooted plant lets at State
Level Biotechnology Centre, MPKV, Rahuri.
