Abstract
Epidemiology of brucellosis in Goshalas of Telangana State was studied using cultural, serological (RBPT, STAT and ELISA), PCR assay (Conventional and multiplex) and milk tests (MRT and mELISA). Out of 550 cattle serum samples screened from two zones of Telangana state, 8.18%, 7.81% and 8.90% were positive by RBPT, STAT and ELISA respectively. An overall prevalence of 7.74% (21), 7.38% (20) and 8.11% (22) was observed in cattle of North Zone of Telangana, whereas 8.60% (24), 8.24% (23) and 9.67% (27) prevalence in South Zone was observed by RBPT, STAT and ELISA respectively. Out of 58 males 4 (6.89%), 4 (6.89%) and 5 (8.62%) and Out of 492 females 41(8.33%), 39 (7.92%) and 44 (8.94%) were positive by RBPT, STAT and ELISA respectively. The prevalence in 1-3 year old was 7.69% by three tests, in 3-6 year old, 8.20%, 7.80% and 9.00% were positive by RBPT, STAT and ELISA respectively, whereas in more than 6 year old, 8.33% by all the three methods. Out of 205 serum samples from human beings belonging to two zones of Telangana states, 14.14%, 12.68% and 15.12% were positive by RBPT, STAT and ELISA tests respectively. The prevalence in the less than 20 years age was 10.52% by all the three tests, in 20-40 years of age, 14.64%, 14.01% and 15.92%, and in more than 40 years of age, 13.79% were positive by RBPT and ELISA and 6.89%, were positive by STAT. Occupation wise the prevalence in veterinarians, 22.22% by RBPT and ELISA, where as 11.11% by STAT, in para veterinary staff, 13.33% by RBPT and STAT and 20.00% by ELISA, in dairy farm workers, 14.20% (RBPT), 13.06% (STAT) and 14.20% (ELISA) and in others 20.00% were positive by ELISA, whereas none was positive by RBPT and STAT, Considering ELISA as gold standard test based on its efficacy, the sensitivity was 85.00% for RBPT and m-ELISA where as 80.00% by STAT and MRT methods. The specificity was 100% for all the methods. Out of 339 milk samples from two zones of Telangana state, 10.32% and 10.91% were positive by MRT and ELISA respectively. For comparing the tests for detection of antibodies in serum and milk,120 samples each of serum and milk were tested, out of which 14.16%, 13.33% and 16.66% were positive by RBPT, STAT and ELISA respectively in serum and in milk 13.33% and 14.16% were positive by MRT and m-ELISA respectively. Cultural, ELISA and PCR assay were compared, where in 10.00%, 15.33% and 12.66% were positive by respective methods, where in milk samples were positive for 9.33%, 16.00% and 13.33% and in blood samples 10.66%, 14.66% and 12.00% were positive by Cultural, ELISA and PCR methods. Out of total 15 isolates of Brucella 11 belongs to any one of 1, 2 or 4 biovars of Br. abortus, 2 belongs to biovar 1 of Br. melitensis and two could not be identified by AMOS PCR.
