Abstract
ticstructurewiththeprimaryobjectiveofscreeningthemaizegenotypes for tolerance to PEG induced stress. Variability was observed in the maize genotypes for morphologicalcharacterssuchasgerminationpercentage,numberofdaysforgermination, root, shoot and total length ofthe seedlings, leaf area, fresh weight and dry weight ofseedlings, root/shoot ratios as well as proline accumulation in root, shoot and leaftissues of 15 days old seedlings. Genotype 98k/5374 exhibited earliergermination, highergermination percentage, rootlength,shootlengthandtotallengthofseedling, leafarea, shootfresh weight and total fresh weight, shoot dry weight and total dry weight followed by 1-31-49. Therefore, it can be concluded that 98k/5374 was lessaffectedbyPEGinducedwaterstress. Germinationpercent, days togermination,shoot,rootandseedlinglength,freshweightanddryweight, leaf area and seedling vigour index decreased as the PEG concentration increased. Proline accumulation was foundto be maximum in leaffollowed bycoleoptile and root tissues. As the PEG concentration increased prolineaccumulation increased signi ficantly in all the three tissues. Genotype98k/5374recordedmaximumprolinecontentinleaftissue,EI-30recorded in coleoptile tissue and EI-Z in root tissue. Least proline content was recordedbyCM-120inleaftissue,CM-120incoleoptiletissueand98k/5711 inroottissue. Callusstudywasrestrictedtoselectedfourgenotypesnamely98k/5374,El-55, EI—4l and CM-120 representing four genotypic groups. 98k/5374recorded maximumrelative callus growth. Proline accumulation in the rootcalli revealed that as the PEG increased there was an increase in prolineaccumulation. 98k/5374 recorded maximum proline accumulation followed by 131-55, while CM-120 recorded minimum proline accumulation. Therelationshipamongvariouscharacterswasfoundtobesignificantlypositively correlated forall the characters studied. Basedongerminationpercent,root,shootandtotallength, leafarea,freshweight,dryweightandseedlingvigourindexof15daysoldseedlings sixteenmaizegenotypesunderpresentstudywere categorised asrelatively resistant, moderately resistant, moderately susceptible and susceptible to drought.
