Abstract
Astudywasconductedwithfourgenotypiegroupsconsistingofl()mar/egenotypesofdivergentgeneticstructurewiththeprimaryobjectiveofscreeningthe maizegenotypesfortolerancetoPEGinducedstress. Intergeneticvariabilitywasfoundinthemaizegenotypesformorphologicalcharacterssuchasgerminationpercentage,numberofdaysforgermination,shoot,root andtotallengthofseedlings,leafarea,freshweightanddryweightoftheseedlingsas wellastheprolineaccumulationinleaf,coleoptileandroottissueof15daysold seedlings. Compositesandsyntheticsingeneralexhibitedhighergerminationpercentage,totallengthoftheseedling, leafareaaswellasfreshweightanddryweightoftheseedlings. AsthePEGconcentrationincreased,theeffectonthesemorphological characterswaslesspronouncedincompositesandsynthetics. Acrossthetreatments,Trishulathaexhibitedhighergemiinationpercentageaswellashighervaluesforallthesemorphologicalcharacters. Effectofhigher concentrationsofPEGontotaldrymatter was of less magnitudeinTrishulathafollowed byRohini,DHM-lOSandVarun. Prolineaccumulationwasfound to bemaximum in leaves followedby coleoptileandroottissue. AsthePEGconcentrationincreased,therewassignilieant increaseintheprolineaccumulationinallthesethreetissues. However,itwasfoundto bemaximuminT3(-6bars) followedbyT2(—3bars)andT4(-9bars). Trishulatha recordedmoreprolineinleaf,RohiniincoleoptileandHarshainroottissuewhile Aswinirecordedleastprolineaccumulationinallthreetissues. Exposureof5dayoldseedlingstodifferentconcentrationsofPEGresultedin considerabledecreaseinshootlength,whilerootlengthwasobservedtoshowvery littledecreaseduetoincreaseinPEGconcentration. Doubleandthreewaycrossesexhibitedminimumreductioninrootandshoot length. Tn'shulathawasfoundtoexhibitstableperformancewhileVaninwasfoundto bemostunstable. Trishulathawasalso foundto resistmorenumberofdays in variousconcentrationsofPEGwhileinbredsCM—l wasearliesttoexhibitsenescence. Numberofdaystakenforsenescenceby 10dayoldseedlingsindifferent PEG concentrationswasfoundtobehighindoublecrossandthreewaycrosshybrids followedbysyntheticsandcomposites. RelativecallusgrowthonrootsegmentswasfoundtobehighinT2(-3bars), thereafteritdecreased. HybridCM—l31xCM-211recordedmaximumrelativecallus growthfollowedbyDHM-lOSandTrishulatha Rohinirecordedminimumrelativecallus growth. ProlineaccumulationinrootcalliwasfoundtoincreaseduetoincreaseinPEGconcentration. RohinirecordedmaximumprolinefollowedbyCM-l3l xCM-2ll and DHM—lOSwhileitwasrecordedtobeminimuminCM-l3l. Therelationshipamongvariouscharacterswasfoundtobesignificantlypositivelycorrelated formostofthecharactersstudied. Totaldrymatterwasfoundtobestronglyin fluencedby totallengthoftheseedlingandleafarea. Heritabilityformostofthecharacterswasfoundtobeveryhigh. Itfacilitateseffectiveselection. ThePEGconcentrationwhich,gaveawaterpotentialof—()bars broughtoutconspicuousandsignificantdifferencef'amongthegenotypicgroupsand individualgenotypeswhichcanbesuggestedforscreeningmaizegennplasmforPEG inducedstress.
