Abstract
ABSTRACT : The objectives of the present study were to ascertain the presence of luteal activity and diagnosis of early pregnancy in crossbred cows. A total of 100 healthy parous cyclic crossbred cows under rural managemental conditions were utilized. Cows were categorized under inseminated and cyclic. In inseminated category 60 crossbred cows on day 22-24 post insemination and in cyclic category 40 non inseminated crossbred cows on day 4, 7 and 17 of the cycle were divided randomly into three groups. In group I, PGF2α 400μg and in group II, PGF2α 600μg injected intrajugularly. While, group III cows served as control without drug. Blood samples were collected from every cow just before PGF2α administration. The response of PGIMFT on day 22-24 post insemination was 68 (17/25), 80 (20/25) and 0.00% (0/10) in group I, II and III, respectively. The time interval for PGIMFT response was 166.47±19.34 and 135.5±16.29 seconds in group I and II, respectively. In group I and II the serum progesterone concentration on day 22-24 post insemination in PGIMFT tested crossbred cows was 9.689±1.25 and 2.796±0.76 ng/ml; and 10.948±1.05 and 3.365±1.05 ng/ml in positive and negative cases, respectively and differed significantly (P≤0.05). The sensitivity and specificity of PGIMFT against the serum progesterone concentration was 75 and 44% in group I; and 89.4 and 50% in group II, respectively and against rectal examination was 73.68 and 50% in group I; and 85.71 and 50% in group II, respectively and serum progesterone against rectal examination was 68.42% and 50% in group I; and 80% and 40% in group II, respectively. In cyclic category the response of PGIMFT conducted on day 4, 7 and 17 of estrus cycle in crossbred cows was 66.67 (10/15), 86.67 (13/15) and 78.57% (11/14) in group I; and 73.33 (11/15), 100 (15/15) and 84.6% (11/13) in group II, respectively. The time interval for PGIMFT response was 187.43±28.79, 145.5±12.29 and 163.93±19.70 seconds in group I and 160.2±15.29, 137.67±21.09 and 153.3±15.29 seconds in group II, respectively. The serum progesterone concentration in group I, on day 4, 7 and 17 of the cycle in PGIMFT tested crossbred cows was 2.751±0.40, 9.546±0.71 and 5.518±0.36 ng/ml in positive; and 0.601±0.35, 6.596±0.45 and 3.593±0.40 ng/ml in negative cases, respectively. In group II, 2.316±0.30, 9.323±0.61 and 5.473±0.33 ng/ml in positive; and 0.558±0.15, 0.00 and 2.665±0.84 ng/ml in negative cases, respectively and differed significantly (P ≤0.05). It was concluded that the test might have a lot of scope for its application in bovine practice for either for making diagnosis of early pregnancy and or detecting presence of luteal tissue.
