Abstract
The research project “Molecular analysis of coconut (Cocos nucifera L.) segregants” was carried out in the Department of Plant Breeding and Genetics, College of Agriculture, Vellayani during 2015 – 2016. The major objective of the study was the Molecular analysis of coconut genotypes developed from controlled hybridization using RAPD and SSR markers. In the study, the selected 120 genotypes of four combinations were subjected to genetic analysis using 5 RAPD and 5 SSR primers. The 5 RAPD primers produced a total of 25 amplicons of which only 8 were polymorphic and giving 32 per cent polymorphism. The amplification product size ranged from 0.5 kb to 2 kb. Among the 5 RAPD primers OPD 3 showed maximum Polymorphic Information Content (PIC). The OPD 3 and OPP 3 primer had highest EMR (Effective Multiplex Ratio) value. OPD 3 primer also produced a specific band to distinguish the parents. Clustering based on Jaccard’s similarity coefficient revealed that all the progenies in the controlled pollination of Komadan mother palms with the pollen collected from the identified Komadan palms were clustered together and shows 100% similarity. In the SSR analysis, all the progenies obtained from the controlled pollinations produced 2 alleles, one from each parent. Among 5 SSR primers, CNZ 43, CnCir H4, CNZ 1, and CAC 50 primers showed maximum value of PIC and EMR of 0.62 and 4.0 respectively. Clustering based on Jaccard’s similarity coefficient revealed that all the progenies in Komadan selfing population had 100 per cent similarity. Population genetic analysis of RAPD and SSR markers revealed that maximum genetic diversity observed in Komadan x CGD population. Molecular analysis with both RAPD and SSR markers showed that, all the progenies in the Komadan x Komadan population had 100 per cent similarity. This indicates the high degree of homozygosity due to predominant self-pollination in Komadan palms.
