Abstract
The present investigation entitled “Shoot tip grafting in Nagpur Mandarin (Citrus reticulata Blanco)” was carried out at the Tissue Culture Laboratory and at the Department of Fruit Science, College of Horticulture and Forestry, Jhalrapatan city, Jhalawar during the year 2011-12. The experiment was laid out in Completely Randomized Design with three replications. Aspects such as rootstock performance, establishment of axenic culture, explants multiplication, in vitro shoot tip grafting and hardening of shoot tip grafted plants formed part of the experiment. The Rough lemon rootstock had the maximum germination (88.66%) and polyembryony (58.61%). The HgCl2 (0.1%) was found as the best treatment as regard to establishment of axenic culture. The maximum aseptic establishment of explants (82.00%) was found when HgCl2 (0.1%) for 11 minutes was used. The maximum number of sprouted shoot (6.23), length of sprouted shoot (2.51cm), minimum no. of days taken to bud break (13.33 days), maximum no. of nodes (5.16) and maximum no. of leaves (7.20) were recorded on MS medium supplemented with 1.0mgl-1 BAP + 0.5mgl-1 NAA (T9). Maximum no. of sprouted shoot (6.67), no. of leaves (5.16) and minimum no. of days taken to bud break (15.00 days) were recorded at 3.0mgl¯1 BAP + 0.5mgl¯1 IBA (T15) and maximum length of sprouted shoot (2.16cm) and no. of nodes (3.76) at 2.0mgl¯1 BAP + 0.5 mgl¯1 IBA (T12). The maximum success of grafts (53.33%) was recorded when 13 days old Carrizo rootstock was used. The maximum graft success percentage (48.33%) was found when 5.00mm size scion was used. With the use of 1.0mgl-1 BAP, the minimum no. of days (10.0 days) taken to bud sprout was observed. However, when the scion was pre-treated with NAA 1.0mgl-1 maximum length (2.00cm) of sprouted shoot was observed. The grafted plantlets had better survival in polyhouse in growing medium consisting of vermiculite: cocopit: perlite: soil in equal proportion. It resulted in graft survival of 91.66% in Carrizo, 86.66% in Rough lemon and 85.00% in Rangpur lime.
