Abstract
The present investigation was undertaken to monitor the genetic differences in some wheat cultivars by carbon monoxide sensitivity of cytochrome c_ oxidase. Some selected wheat cultivars were also tested for CO sensitivity with coldpre-treatroent at 4°C for 18 hrs, and effect of TCA cycleintermediates on CO reaction. Gibberellin CGA-rD sensitivityof different wheat cultivars and effect of cold pre-treatment C4°C tor 20 hrs.3 on 6A3 sensitivity were also studied.Seedlings of wheat plants were grown in soil filled earthen pots under natural day-light at the Department ofBiochemistry, Manatma Phule Krishi Vidyapeeth, Rahuri, during1992 and 1993, for screening of wheat cultivars for C0-sensitivity of cytochrome c_ oxidase. Gosavi The Leaves of 10 day-old seedlings were used to screen carbonmonoxide sensitivity by measuring nitrite production during in vivo nitrate reduction assay under aerobic and anaerobicconditions.In CO aerobic assay of nitrate reductase after one min exposure to CO, wheat cultivars were found to be completely insensitive to carbon monoxide, indicating that thecytochrome a-* in these cultivars was largely in the oxidized state. Six cultivars of wheat [without and with Rht genes]were tested for their CO sensitivity at the defined CO:02ratios. A higher concentration of CO was found to benecessary for the formation of cytochrome a^-CO complex. The cold pre-treatment at 4°C for 18 hrs enhanced the CO response. When the leaves of wheat plants were infiltrated with TCAcycle intermediates at 10 mM concentrations, the C0-sensitivity of cytochrome c_ oxidase was enhanced.s The four wheat cultivars were tested forgibberellin CGA-rl sensitivity at 25°C for 48 hrs as well as at4°C for 20 hrs. The tall wheat cultivars were found to besensitive to 6A^ at the ambient temperature. However, thedwarf wheat cultivars which were found to be insensitive toGA-r at the ambient temperature becames sensitive to GA-^, afterthe cold pre-treatment at 4°C for 20 hrs.
