Abstract
Two varieties each of two cultivated species and four wild species were subjected to biochemical and molecular mvestigations in order to understand the boUworm complex tolerance m cotton The studies involved collection of healthy and spotted, American and pink boUworm infested samples of leaves, squares and bolls at 58, 66 and 98 DAS of the varieties of cultivated and wild species It was observed that mean trypsin inhibitor activity m leaves, squares and bolls was 48.30, 45.52 and 44.22 units ml"1, respectively in wild species and was significantly higher in the corresponding plant parts of the varieties of cultivated species. Comparatively higher polyphenol contents were observed m the leaves, squares and bolls of wild species than m the susceptible cultivated species The contents of structural barriers like ADF, NDF, cellulose, hemicellulose and lignrn were correspondingly at XV11 Abstract Contd SL Patil higher level m the above plant parts of wild species than the vaneties of cultivated species. However, the soluble protein contents were significantly low in above- plant parts of wild species than the aneties of cultivated species. The PPO and POD activities were of higher magnitude in plant parts of wild species than the vaneties of cultivated species The leaf soluble protein profile depicted that the protein bands of 51.28, 22.38 and 20.89 kD size as resolved by SDS-PAGE and 56 23 and 32.35 kD as resolved by Native-PAGE were present only in wild species However 63.09, 56 23 kD size as resolved by SDS-PAGE and 53.70, 48.97 and 19 95 kD size as resolved by Native-PAGE were present only in vaneties of cultivated species. Out of a total of 8 primers employed, only three exhibited polymorphic bands The pnmer OP A-11 synthesized the unique bands of the size of 1.58 and 1 25 kb only m resistant wild species The higher levels of trypsin inhibitor activity, polyphenols, ADF, NDF, cellulose, hemicellulose and lignm contents and lower levels of soluble protein in resistant wild species of cotton and the inverse proportion of these factors resulted in spotted, Amencan and pink bollworm attack at 58, 66 and 98 DAS as appeared in vaneties of cultivated species The unique polypeptides appeared in cultivated or wild species may have their association with susceptibility or resistance to bollworm complex and can be used in marker assisted screening for resistance to these pests in cotton. The unique bands of 1.58 and 1.25 kb size synthesized by OP A-11 pnmer only in wild species is probably linked to the resistance to bollworm omplex
