    Genetic variability, path analysis and stability parameters in sesame

    Biometric analysis in a varietal collection of sesame was undertaken to study the genetic variability, correlations, path analysis and stability parameters. One hundred sesame types were evaluated in replicated trials at Vellayani in uplands during rabi and at Kayamkulam in rice fallows during summer. Genetic variability and correlations were estimated and path analysis worked out independently as both the locations. Location trials for estimating stability parameters were conducted at three places viz. in uplands during rabi at Pattambi and Vellayani and in rice fallows during summer at Kayamkulam. Large values for genotypic coefficients of variation were obtained for characters such as number of capsules on branches, number of capsules perplant, number of capsules on main stem and number of branches during rabi as well as summer. The lowest genotypic coefficient of variation was obtained for number of days to maturity during both rabi and summer. High values of heritability were recorded by seed protein content , seed oil content, height upto first capsule and weight of 1000 seeds under both conditions.

    Influence of applied nutrients and stage of harvest on the yield and physicochemical properties of essential oil of palmarosa (Cymbopogon martini Stapf var. motia)

    Palmarosa, an essential oil crop introduced in Kerala, from Maharashtra, nearly two decades ago, is spreading steadily la the pialas and midland regions of North Kerala. Several agro-techniques have yet to he standardised for the commercial cultivation of this crop in the State. The present studies were undertaken at the Aromatic pnfl Medicinal Plante Research Station, Odakkaly during 1980-'8 4 to obtain information on the nutrition of palmarosa, optimum harvest intervals to ensure maximum herbage and oil yields and the factors influencing the quality of oil. The treatments in the major field experiment (1980-'82) consisted of three levels of N, and KgO, each at 25, 50 And 75 kg/ha along with six intervals of harvest at 40, 45, 50, 55, 60 and 65 days. The total number of treatment combinations wore 162 in a 3^ x 6 confounded asymmetrical factorial design. Tho main experiment was continued for another two yoaro limiting tho observations, to the yield of herbage and oil. This was then followed by an observation trial with intervals of harvest longer than tho maximum of 65 doye tried in the main experiment. In the main experiment the herbage yield vns significantly increased by application of P2°5 ^2°* Nitrogen did not have any eignifioant offeot on herbage yield poaeibly due to tha medium level etatua of soil N in the ABSTRACT ^erimental plots. The oil yield was Bignifioantly enhanced by P205 application at 50 kg/ha. Different levels and K showed no significant influence on the yield of oil. Harvest intervals showed significant influence on herbage yield, oil yield and oil content and the maximum value was recorded by the 65 day interval. A path analysis of the herbage yield with related cnaracters has shown that the height of the plant is the most important yield attribute influencing directly the yield of herbage. Path analysis of oil yield showed that oil yield is mainly dependent upon herbage yield. The direct effects on oil yield by yield attributes are found to be in the decreasing order of number of tillers with inflorescence, height of the plants and length of inflorescenoe. Maximum indirect affect via herbage yield is expressed by the height of the plant and length of inflorescence. All these directly and indirectly contributing factors arc soon to be markedly influenced by tho applioation of phosphorus and intervals of harvoat* Herbage yield and oil yield for various harvest lntorvalo obtained In the flrot two years wore fit tod In a Cobb-Dcughlua reoponoo function and tha expooted values «leulat«d were found to be very olooe to tho actual observed values• In thethird and fourth year of the experiment, the ghest herbage and oil yields were recorded by 60 day interval followed by the interval of 65 days, A quadratic Sanction for the data pooled over for four years was fitted to see if the data show a diminishing return with an increase in harvest interval and it was found that the optimum herbage yield and oil yield were obtained when the harvest was done at 62 and 63 days interval respectively. The final observational trial also indicated that the herbage and oil yields were maximum for a harvest interval of 65 days beyond which it decreases. Increase in the levels of both N and P tended to increase the content of geraniol and to decrease the content of geranyl acetate which are the price determining quality attributes of the essential oil. But the maximum interval of harvest viz., 65 days tried in the main experiment was not sufficient for moot of the physico-chemical properties of oil ouoh os specific gravity, refractive index, geraniol content, geranyl acotato content etc. to reach the minimum limit prescribed by 131. The observational trial showed that the oil obtained at intervale of 95 days and above satisfied the 131 specification with respect to all the phyalco-ohemioal properties of the oil. However, at thle herveat Interval the yield Itself wae considerably depressed compared to the yield at 65 day Interval. Th, eoonomloe worked out for different Intervale of harveab baaed on the ourrant market prloe of the oil paeein« as above the I SI limits as fe. 240/- per kg (oil from harvest intervals at and above 95 days) and those below them at Fs.220/- per kg (oil from harvest intervals of 55, 65, 75 and 85 days) it has been found that a wider cost benefit ratio and net return per rupee investment are obtainable for harvest intervals ranging from 65 to 85 days. However, 65 day interval has the advantage of early returns from the investment. Application of 50 kg PgO^/ha in view of its significant effect on herbage yield, oil yield and oil quality increase the net profit per rupee invested for the harvest interval of 65 days. The average removal o f II, P, K, Ca and Mg from the s o i l by palmarosa per h ecta re per year lias a lso been worked o u t .     Dietary protein and energy requirements of caged layers

    A 4 x 4 factorial trial designed to study the dietary requirements of protein and energy for caged egg type layers under the hot - humid conditions of Kerala is presented in this thesis. Single Comb White Leghorn strain cross pullets (IWN x IWP ) of the Mannuthy centre of AICRP on Poultry for eggs formed the experimental subjects. The birds were randomly alloted to 16 dietary protein-energy combination groups. The dietary protein levels employed were 14,16,18 and 20 per cent and that of energy levels were 2400, 2500, 2600 and 2700 Kcal ME per kg diet. General observations of the experiment were recorded over 12, 28 day periods. The varying levels of protein and energy employed in this study did not have any significant influenced on body weight gain. Birds fed 18 per cent dietary protein had significantly higher hen-day egg production and it was significantly lower for birds fed 14 percent dietary protein. Egg production observed between 16 and 20 per cent and that between 18 and 20 per cent protein were not statistically significant. Hen-day egg production was not influenced by various dietary energy levels employed. The daily feed intake per bird per day was not affect by the le4vels of protein and energy employed in this experiment. A dietary protein levels of 16, 18 and 20 per cent showed significantly superior feed efficiency, when expressed both in terms of feed per dozen egg as well as feed per kilogramme egg. Feed conversion efficiency was not influenced by verying energy levels employed. The egg weight was significantly influenced both by protein and energy levels. However, the changes exerted due to feeding of various dietary combinations did not show any definite trend. The ,mean shell thickness was significantly influenced by dietary protein levels whereas energy did not have any effect. There was a statistically significant decrease in albumen index and Haugh unit score and albumen index was influenced significantly by the energy level in the diet. The yolk index decreased with increase in protein content in the ration, whereas it was inconsistent with the dietary energy levels. Biochemical parameters such as haemoglobin, serum protein, serum inorganic phosphorus and packed cell volume were not influenced either by protein or energy levels. The level of dietary protein did not influence the serum lipid levels whereas the dietary energy had influenced. The dietary- protein-energy combinations in the diet significantly influenced liver lipid and liver lipid and liver protein. The values obtained on carcass analysis were well within the normal biological limits and did not show any specific effect due to different dietary treatments . All the birds showed a positive nitrogen balanced irrespective of the protein or energy levels in the diet. On the basis of these results it can be inferred that the levels of dietary protein and energy required for optimum production for layers raised in cages are 16 per cent and 2400 Kcal ME per kg, respectively. However, in practical feed formulation where amino acids balancing especially that of the critical amino acids being a problems in developing countries it is suggested that a dietary protein level of 18 per cent can be considered as optimal.     Selection studies in chicken for egg number on part records

    All India co-ordinated Research Project on Poultry Breeding for egg located at Kerala Agricultural University, Mannuthy , Trichur is maintaining two strains of white Leghorn which are subjected to selection for high egg production since 1978. The criterion of selection has been IDS method of index selection by combining the information on sire and dam family averages with the individual’s performances, for egg number up to 280 days of age. The present investigation was planned with the following objectives. -To evaluate the response of intrapopulation selection for part period egg production. -To estimate the genotypic and phenotypic parameters for different traits. -To evaluate the the correlated responses in unselected traits. -To measure the time trend in genetic parameters. -To measure the interstrain variation for some economic traits. To achieve these objectives data collected over a period of five generations, including base generation, on the experimental populations were utilized. Chicks hatched during 1979-80 hatching year of each strain constituted the foundation stock (S0). Thereafter both strains were subjected to four generations of selection. Standard managemental throughout experimental period. practices were followed uniformly The traits measured were egg number up to 280 day of age (EP), age at first egg in days (ASM), body weight in grammes at 20 and 40 weeks of age (20 wk-40 wk BW ) and egg weight (BW) in grammes at 30.40 weeks of age. The average effective number of male breeders, female breeders and progenies with complete records were 40, 238.4 and 1165.8 in IWN and 39.8, 232.2 and 1076.6 in IWP strains, respectively, over generations. The effective populations size over generations were 136.62 in IWN and 135.33 in IWP strains. The coefficient of inbreeding increased by two per cent in both populations over four generations of selections which would be the upper limit due to intentional avoidance of sib matings. Since hatch effects were significant for most of the traits, data were corrected for hatch effects before analysis by fitting least square constants. The heritability of each trait and correlations between traits were estimated initially intra generation-strain basis following variance and covariance component anaylsis and then pooled over generations within strain to provide mean estimates. The heritability estimates of various traits obtained in this study were consistent with several earlier reports. Sexlinked effects appeared to be important in the inheritance of EP 20 WK, BN, EW and ASM in IWP strain whereas maternal effects wee important for 40 WK, BN. But sex-linked effects for 40 weeks BN appeared to be important in IWN. The genetic and phenotypic correlations of the present study were close to several earlier reports both in direction and magnitude. The genetic parameter estimates did not show any significant time trends due to four generations of selection in both populations. The realized phenotypic response per generation obtained from regression of generation means on generation number were 8.30 + 1.60 eggs in IWN 5.37 + 1.6-0 eggs in IWP strains, which were significantly different from zero. The correlated response per generation for ASM, 20wk BN, 40 wk BN and BN due to selection for part period egg number were-4.26 d, 28.46 g, - 10.32 g, -0.28 in IWN and –4.45 d, 37.28 g, -13.80 g, -0.02 g in IWP strains, respectively. The realized gains in selected and unselected traits agreed well with the predicated response in direction but not in magnitude, except 40 wk in IWP which was in opposite direction. For all the traits the realized gains were found to be higher than the predicted gains except BN of IWP which was less than the predicated response. Difference were highly significant between the two strains for all the traits under study in the generation except S2 for BN : S0 for ASM, 20 wk BN: S3 for 40 wk BN : S0, S1 and S3 for BN.

    Calcium and phsophorus requirements of caged layers

    A 4 x 4 factorial arrangement to determine the dietary requirements of calcium and phosphorus for caged layers under the hot-humid agro-climatic conditions of Kerala is presented in this thesis. Two hundred fifty six single comb White leghorn strain cross pullets (IWN x IWP ) of the Mannuthy Centre of AICRP on Poultry foe eggs formed the experimental birds. The birds were randomly assigned to 16 dietary calcium-phosphorus combination groups. The dietary calcium levels employed were 2.50, 3.00, 3.50 and 4.00 per cent and that of phosphorus levels wee 0.50 , 0.60, 0.70 and 0.80 percent the observations of the experiment were recorded over 10,28 day – periods The percent hen-day egg production and feed efficiency (both on egg number and egg mass) were significantly (P < 0.01) superior in birds fed 4.00 percent dietary calcium. The dietary levels of phosphorus had not exerted any influence on hen-day egg production and feed efficiency. The feed intake and egg weight were not influenced either by the dietary levels of calcium or phosphorus or both. The egg shell quality (specific gravity and shell thickness ) was significantly (P < 0.01 ) better in egg laid by hens fed 3.50 and 4.00 percent dietary calcium. The percent egg shell was significantly (p< 0.01) highest with 3.50 percent calcium. However, percent egg shell between 3.50 and 4.00 percent dietary calcium was not statistically different. The egg shell quality (specific gravity, shell thickness and percent shell) was significantly (P< 0.01) poorest with 2.50 percent calcium diet. The dietary phosphorus levels had not exerted any significant influence on egg shell quality (specific gravity, shell thickness and percent shell). The serum calcium level, serum inorganic phosphorus level and percent tibia ash were not influenced by the levels of dietary calcium and phosphorus . The serum alkaline and acid phosphatase levels did not give any clue as to the adequacy of the dietary calcium and phosphorus level employed. The calcium intake was higher with higher levels of dietary calcium. There was a trend of increased absolute calcium retention as the calcium level in the diets increased but it was not statistically significant. Dietary phosphorus levels positively improved absolute retention calcium. The mortality among birds were nit suggestive of any influence due to dietary calcium and phosphorus. The results of experiments revealed that in terms of any of the major economic traits used for evaluating the dietary calcium adequacy of caged layers, 4.00 percent calcium was found to be superior over the other levels. In respect of dietary phosphorus, the levels of 0.50 per cent seems to be adequate but the absolute retention of phosphorus was negative at this level of incorporation irrespective of calcium levels. Therefore, it will be more logical to set the requirements of dietary phosphorus at 0.60 per cent than at 0.50

    Selection efficiency and genetic and biochemical bases of resistance to bacterial wilt in tomato

    Bacterial wilt (Pseudomona solanacearum E.F. Smith) is a major disease of tomato. Attempts were made to improve a reportedly resistant line CL 32d-0-1-19 GS for higher fruit weight and better plant type through four selection methods – mass, pureline, single seed descent and bulk. Genetic and biochemical bases of resistance were also studied. Resistance of the evolved line was tested in vivo and in vitro. Mass, pureline and SSD methods of selection were effective to improve fruits/plant, locules/fruit, yield/plant and fruit weight. SSD method resulted in higher realized genetic gain for fruits/plant (30.97) locules/fruit (0.5) and yield/plant (1.05 kg). Higher genetic advance and high realized heritability were recorded for days to first harvest in SSD selections. Mass selection had higher realized heritability (0.95) for fruits/plant and high realized heritability (0.65) and realized genetic gain (0.5) for locules/fruit. Fruit weight was improved through pure- line selection. Selections based on trait combination fruits/plant, yield/plant and plant height significantly effected higher locules/fruit (4.2). Selection based on trait combination fruits/plant, yield/plant, locules/fruit and plant height were the earliest to flower (59 days). Days to first harvest were significantly reduced (96 days) through selections based on fruit/plant and yield/plant followed by yield/plant and plant height (98 days). Fruit weight was significantly improved by selections based on yield/plant and locules/fruit (58.5 g). Selections based on yield/plant had high realized heritability (0.59) and realized genetic gain (22.0 g) for fruit weight. Fruit weight was significantly improved by selections based on yield/plant and locules/fruit under SSD method (62.4 g). Transgressive segregants appeared through selection based on yield/plant and locules/fruit. SSD selections had the lowest incidence of wilt (9.86%). Multi- locational trails revealed a survival of 89.5% of plants under normal disease stress. Evaluation of generations from LE79 (CL 32d-0-1-1-1-19GS) x Pusa Ruby Cross indicated a monogenic and incompletely dominant type of gene action for wilt resistance. There was no association between yellow gel colour around the seed and disease resistance. No close linkage between resistance and a fewer locules/fruit was observed. The resistant line (LE79-CL 32d-0-1-1-1-1-19 GS) had higher total and higher root content of Pusa Ruby. - tomatine than the susceptible line - Tomatine content increased and maintained at a higher level in resistant line eventhough the infection progressed consequent to artificial inoculation. Pusa Ruby wilted seven days after inoculation. Total phenol content was higher in roots of Pusa Ruby before and after inoculation. The wilted plants of Pusa Ruby had higher content in root and shoot. The O.D. phenols content was more in the resistant line before and after inoculation. Vitamin C content was also more in roots of LE79 before and after inoculation. A higher ratio of phenols and -tomatine: total phenols, -tomatine: O.D. -tomatine: vitamin C were found in roots of LE79 before inoculation. The wilted plants of Pusa Ruby had lower ration of -tomatine: total phenols and -tomatine: vitamin C. a higher increase in O.D. phenols and vitamin C content on infection was observed in resistant line. The ratio of vitamin C: total phenols was higher in roots of LE79 before infection and the ratio increased in both the lines initially on infection and then decreased but to a greater extent in Pusa Ruby. A higher increase in vitamin C content compared to total phenol was observed in LE79 on infection. A low ratio of total phenol: O.D. phenol was related to resistance in LE79. Inoculation in vitro confirmed the resistance of LE79 to Vellanikkara isolate of Pseudomonas solanacearum E.F. Smith. Grafting of suspectible scion on LE79 delayed wilting of scion even on artificial inoculation.
