ThesisItem Restricted GENETIC EVALUATION OF SEMEN PRODUCTION AND FERTILITY TRAITS IN EXOTIC, CROSSBRED AND INDIGENOUS CATTLE AND BUFFALOES(2021) BHAVE KAUSTUBH GAJANAN; TANUVAS; THILAK PON JAWAHAR K; KUMARASAMY P; SIVAKUMAR T; CECILIA JOSEPH; ENKATARAMANAN RThe records of 736 bulls comprised of 2,69,795; 9,662 and 2,239 ejaculates of semen production, semen quality and sperm abnormality data respectively, while, 5,76,870 artificial inseminations on different genetic groups were broadly classified into five categories viz. Holstein Friesian, Jersey, crossbreds of Holstein Friesian and Jersey, indigenous cattle and buffaloes. The effect of genetic and non-genetic effects on semen production, semen quality, sperm abnormality, and fertility traits were tested using linear mixed model. The effect of genetic and non-genetic factors on lifetime semen production performances were analysed using general linear model. The variance components were estimated by REML method using the animal repeatability model (univariate and multivariate) while, the breeding values were estimated using BLUP method. The order of ejaculate, collection interval, time of collection, breed × age at collection, semen collector × year of collection and month × year of collection had a significant influence (P<0.01) on ejaculate volume, sperm concentration, total sperm, mass activity (only for buffaloes), initial motility, total motile sperm, post-thaw motility, and total motile sperm after thawing in both cattle and buffaloes. In case of sperm abnormality traits, level exotic inheritance, age at collection, had no effect (P>0.05) on Jersey crossbred bulls, while, the age at semen collection had a significant (P<0.05) effect on only mid-piece abnormality, total abnormal sperm, and total normal sperm of Holstein Friesian crossbred bulls. The breed and age at semen collection had a significant effect (P<0.05) on semen quality traits involving acrosome integrity of neat and frozen semen, hypo-osmotic swelling test and incubation tests in cattle. While, the breed and age at semen collection were mostly non-significantly influenced semen quality traits in buffaloes. However, season × year of collection significantly (P<0.01) affected (P<0.01) both sperm abnormality and semen quality traits. The fertility trait was significantly affected (P<0.05) by body condition score, stage of heat and order of parity etc. on different genetic groups.ArticleItem Open Access Medical termination of pregnancy in bitches using progesterone receptor blocker(2006) KULASEKAR K; CECILIA JOSEPH; RANGASAMY S; TANUVASThis trial was undertaken to test the efficacy of progesterone receptor blocker, Mifepristone (Tab.Mifeprin-200 mg, Sun Phannaceuticals, Mumbai) in terminating pregnancy in bitches. Ten pregnant bitches brought to the Madras Veterinary College Hospital for medical termination of pregnancy were administered Mifepristone at the dose rate of2.5mg/Kg body weight bid orally for 4-5 days. Ultrasonography was done to confirm pregnancy and to monitor fetal viability and resorption after induction of treatment. In all bitches, pregnancy was terminated between 3-6 with a mean of 4.5 ± 1.5 days after treatment and there was no side effect associated with the treatment. Mifepristone administration is a safe and reliable method to terminate unwanted pregnancy in dogs.ArticleItem Open Access Prevalence of reproductive conditions in canines(2005) CECILIA JOSEPH; KULASEKAR K; AMRITHA ARAVIND; THILAGAR s; TANUVASThe prevalence of reproductive conditions in canines were analysed from 2498 clinical cases presented to the Small Animal Gynaecology and Obstetrics Unit of Madras Veterinary College hospital during the year 1999 to 2001. The incidence of physiological conditions (61.9%) like breeding advice and pregnancy diagnosis were higher than the pathological disorders (38.1%) like pseudopregnancy, anestrum, medical termination of pregnancy, pyometra, dystokias, postpartum conditions and venereal tumors. Higher incidences of reproductive conditions were recorded during South West Monsoon (27.94%). Majority of the reproductive problems occurred in bitches aged 1 to 2 years (42.31 %) and in the Spitz breed (38.71 %).ArticleItem Open Access PROINFLAMMATORY CYTOKINE EXPRESSION AS A EARLY DIAGNOSTIC MARKER FOR DETECTION OF SUB CLINICAL ENDOMETRITIS IN REPEAT BREEDER CROSSBRED COWS(2015) KALYAAN US; KULASEKAR K; CECILIA JOSEPH; et al.; TANUVASA clinical study was carried out in a group of crossbred cattle with the history of repeat breeding. Ten apparently healthy cows having a normal calving history and clear estrual mucus discharge were selected as Control (Group I).Ten repeat breeder cows with watery,cloudy, copious discharge suspected for sub clinical endometritis (SEND) were selected for the study (Group II). These cows were subjected to gene expression studies of pro inflammatory cytokines IL - 8 and TNF a from endometrial biopsy samples using qPCR and their expression was found to be increased compare to control groups, thus indicating that pro inflammatory cytokines such as ILB and TNF a can be used as a diagnostic indicator for SEND.ArticleItem Open Access RECOVERY OF CANINE OOCYTES BY SLICING METHOD(2013) UMAMAGESWARl J; CECILIA JOSEPH; KULASEKAR K; et al.; TANUVASOvaries were collected from bitches undergoing routine ovario-hysterectomy and transported in physiological saline at 35°C to 38°C within 2 to 3 h of collection. A total of 1616 oocytes were retrieved by slicing of 52 ovaries with a recovery rate of 31.07 ± 3.47 oocytes per ovary. Among the collected oocytes 25.93, 43.07 and 31.00 percent of oocytes were graded as I, II and Ill respectively.
