Title: STUDIES ON THE CHARACTERIZATION AND IDENTIFICATION OF GENOTYPES BASED ON MORPHOLOGICAL, BIOCHEMICAL AND MOLECULAR MARKERS IN HORSE GRAM (Macrotyloma uniflorum (Lam.) verdc.)

Abstract: The ability to distinguish and clearly identify varieties of cultivated species is fundamental for the operational aspects in the seed tread. A comprehensive study was conducted during 2008-2010 on the characterization and identification of genotypes based on morphological, biochemical and molecular markers in horse gram (Macrotyloma uniflorum (Lam.) verdc.). The salient findings of the study are detailed below. The horse gram genotypes were classified into six groups as Brown, Light brown, Reddish brown, Yellowish red, Black and Very dusky red based on seed colour. CRHG- 4, CRHG-18, TCR-614 genotypes were grouped as black whereas; BGM-1, BJPL-2, CRHG-17, HG-34 and PHG-9 were grouped under brown colour while VLG-22 and TCR-74 genotypes were grouped under Reddish brown. But AK-42, AK-42C, PHG-9C, VLG-21, TCR-223 genotypes were grouped under yellowish red colour while only AK- 38 had very dusky red colour on seed. Similarly, horse gram genotypes were grouped based on colour of streak around the hilum viz., black, light brown and dark brown. The black hilum colour was noticed in AK-42C, CRHG-4, VLG-22, TCR-81, TCR-73, TCR- 268, TCR-614, TCR-74, AK-42, PHG-9 genotypes. While, BGM-1, CRHG-17, CRHG- 18, PHG-9C, VLG-21, TCR-117, TCR-244, AK-38 genotypes revealed light brown hilum colour and rest of the genotypes were dark brown in colour. Based on green colour intensity on leaf, the genotypes were classified into (light, medium and dark colour). Genotypes AK-42, BGM-1, BJPL-2, CRHG-4, CRHG-18, PHG-9C, TCR-117, TCR-268, TCR-614, TCR-74 and PHG-9 were grouped under medium green colour whereas, AK-42C, CRHG-17, HG-34, VLG-22, VLG-21, TCR-244 and AK-38 exhibited dark green and others light green colour. Based on pod length, the genotypes were grouped into three categories (short, medium and long) genotype BGM-1, BJPL-2, HG-07, TCR-81, TCR-117, TCR-244, TCR-268, TCR-614 and TCR-74 were grouped under long category (>6 cm) whereas VLG-22 and TCR-73 were classified under short category (<5 cm) and rest of the genotypes under medium category (5-6 cm). The electrophoresis separation of soluble seed protein zymogram indicated that, most of the tested cultivars could be easily distinguished in the Region B (66.0 to 97.4 kDa) , C (55.6 to 66.4 KD) and E (27.0 to 42.7 KD) since the banding pattern was quite distinct for each genotype in these regions. Similarly Esterase and Peroxides isozymes also produced marked differences in the banding pattern and their intensities could be used for the identification of horse gram genotypes. Further, set of 10 SSR primers amplified 26 alleles from 22 horse gram genotype. On an average of 2.6 alleles per marker were amplified. All most all the primers amplified minimum of 2 alleles. Some of the primers viz., CA910489, CA910598, CA911600, CA911990 and CA912170 amplified maximum of 3 alleles. Highly distinct genotypes among the selected germplasm were BGM-1 with TCR-74 and AK-38 followed by TCR-74 with TCR-614 and VLG-22 with TCR-223 which could be successfully utilized in the breeding programme of horse gram crop improvement. The study has generated some important morphological traits, protein marker and molecular markers that can be effectively employed in distinguishing the horse gram genotypes for the purpose of DUS testing.
